Technical Background
ACE-031 is a soluble ActRIIB-Fc fusion protein (~101–120 kDa) composed of the extracellular domain of activin receptor type IIB linked to a human IgG1 Fc fragment. It functions as a myostatin trap, binding myostatin, activin, and related ligands with high affinity. The Fc component extends circulating half-life to approximately 10–15 days, allowing for sustained systemic exposure. Clinical trials in Duchenne muscular dystrophy demonstrated trends toward preservation of lean body mass but were discontinued due to adverse events.
